This article is cited by 16 publications. This work provides insight into the structural diversification of aromatic polyketide natural products via unusual redox tailoring reactions that appear to defy biosynthetic logic. While RslO9 can further convert the rearranged intermediate to rishirilide D and lupinacidin A, an additional ketoreductase RslO8 is required for formation of the main products rishirilide A and rishirilide B. Flavin monooxygenase RslO9 then oxidatively rearranges the carbon backbone, presumably via lactone-forming Baeyer–Villiger oxidation and subsequent intramolecular aldol condensation. First, the flavin-dependent RslO5 reductively ring-opens the epoxide moiety of an advanced polycyclic intermediate to form an alcohol. In this work, we employ gene deletion experiments and in vitro enzyme studies to identify key biosynthetic intermediates and expose intricate redox tailoring steps for the formation of rishirilides A, B, and D and lupinacidin A. ![]() In rishirilide biosynthesis by Streptomyces bottropensis, 13C-labeling studies previously implied extraordinary carbon backbone and side-chain rearrangements. The structural diversity of type II polyketides is largely generated by tailoring enzymes.

What we need is meaning in life, which then leads to happiness.Īt Tohoku University in Japan, a 7-year study of 43,000 adults aged 40 to 79 were asked if they’d had a meaningful life, and then tracked what happened next. Don’t Confuse Instant Happiness with Long-Term Happiness When we schedule something, we’re more likely to do it, because the decision to do it has already been made. Love a yoga class but often lack the time? Find the time and put it in your calendar. What to do? Regularly schedule happiness appointments. Just waiting for happiness to come is not a recipe for success. We value happiness so much, but when we’re happy we usually put it down to good luck. ![]() Not spending enough time with the people we love is the one thing we regret the most later in life. If you’re feeling a bit blue today, spend time with friends. Indeed, the capacity to love, and be loved, is a single strength most clearly associated with subjective well-being at age eighty. Indeed, the researchers of the GRANT study wrote that “connecting with and helping others is more important than obsessing over a rigorous exercise program”. This finding was not only limited to university students but in the GRANT study, which followed men throughout their entire lives. Good relationships were a stronger predictor of happiness than student’s university grades, family income, SAT score, age, gender and ethnicity. The happiest 10 percent of Harvard Undergraduates reported having strong social support. There is one characteristics which is the most prominent in happy people – strong social ties. With all this being said and done we need to discover the various aspects that made the operating system apart from the other line up of operating systems. this boosted up sales and also brought fame to the line-up of the Mac OS X series of operating systems. With the release of the Mac OS X Tiger, the software industry felt threatened because of the high-end performance of the system. they started releasing a range of great operating systems, of which the Mac OS X tiger holds the record of being in use the longest. Apple released an operating system series called OS X. now you can say that all systems have a limit, but the performance of these systems comparatively is very good and the creditability of the systems is on par. One very feature is the smooth transition while operating the system. There are a lot of aspects that make these operating systems powerful. ![]() You can go to the Downloads section, scroll down to the OS X category and try and download any of the previous versions.Mac Operating systems are definitely by far the best-operating systems in terms of features and reliability on those features. I'm having troubles downloading the software, I left a message on Aegisub forum but no solutions yet. It is very different from SW in how it works, however, so you will have to adapt. Bear in mind, however, that it was created specifically for anime fansubbing - and it excels in that domain, but when it comes to other kinds of subtitling, it does not provide sufficient functionality (for instance, it has no checks save for reading speed and line length).įor the time being, you should use Aegisub it is definitely one of the best (if not the best) free subtitling tools compatible with Mac. I'll be brutally honest here: Subtitle Workshop doesn't cut it for professional subtitling - for many reasons, but mainly because it lacks shot change detection, which makes creating accurate, standards-compliant timecodes simply impossible.Īegisub is a superb tool that I used for subtitling for 4 years. But I work with a Mac, so I'm trying to find, if possible, a free software for the time being. I'm a translator but wanted to learn more about subtitling and I'm just starting an online course with a university in the UK which uses Subtitle Workshop. ![]() In some cases it is worth typing -W to avoid the extra connection attempt. And every one deserves a retirement at some point Plus, a new developer could work on porting TC to other platforms. However, as I mentioned, perhaps bringing in a fresh developer into the mix would be a good idea for 2 reasons. Unfortunately, I can't see a reasonable business plan that would allow the developer to do that. I wish there was some sort of way for Ghisler to open source this project and still continue to make money from it, and others could do the work of porting it to Linux and Mac. ![]() Free Pascal is designed with the philosophy of "write once, compile anywhere." I understand that in practice it's not nearly as simple, but overall it would be possible to port TC to both Linux and Mac with some effort. I recently learned that TC was written, or rather, ported to Lazarus and Free Pascal. Second, in general Mac Users are just more likely to actually pay for software.Īny thoughts? I know it's not easy to crosscompile apps for 3 operating systems, but I also think that it would be profitable and beneficial to the world. First, because all OSX users would be new customers, not old customers simple downloading updates for free. (just and idological thing, not because they are deadbeats, I am a linux user my self) However, OSX side might in fact be very, very profitable for Ghisler and company. The Linux side of things would not be very profitable, as Linux users in general do not like paying for software. Also, there are frameworks available for Delphi to also make it cross compilable.Īs a result, I was wondering if Ghisler might be interested in perhaps cooperating with another developer, perhaps some one skilled in OSX and Linux coding, to port the Total Commander first to a crosscompilable framework, and then to OSX and Linux. Total Commander is written in Delphi 2, which is a derivative of Pascal. However, I did notice that Double Commander can be compiled cross-platform due to it's use of Free Pascal toolkits. The Double Commander is a grate little clone of Total Commander, but the development on it is so painfully slow, and at the moment it's just much too buggy. ![]() Out of those the only solid alternative at the moment is muCommander. I love the Operating system, and even more so, I love the hardware. I have recently made the switch to mac, and for the most part I am very happy. I know this has been proposed and rejected in the past, but I know there is a serious need and desire for total commander on mac and linux operating systems. |
